Mechanism of Action

Long-R3-IGF-I is a mutein of insulin-like growth factor I carrying a 13-residue N-terminal extension and an arginine substitution at position 3. A 1996 study in Growth Factors designed and characterised these muteins specifically to reduce affinity for IGF binding proteins, which in native IGF-I sequester the great majority of circulating peptide.

Solution NMR work published in the Journal of Biological Chemistry in 2000 described the structure and backbone dynamics of the long-[Arg3] variant, providing the structural basis for the altered binding-protein interaction while receptor recognition is retained.

Research Findings

Comparative animal work published in the British Journal of Nutrition and in Domestic Animal Endocrinology examined growth and protein metabolism endpoints in artificially reared neonatal pigs and in beef heifers. These agricultural studies are the most direct published characterisation of this specific analogue.

The wider IGF-I literature includes work on receptor activation in mammary tumour models, which is relevant safety context for any protocol involving IGF-I receptor agonism. Human data on this analogue specifically are not indexed.

Storage & Reconstitution

Sealed lyophilized vials are stored in the dark. The peptide-formulation literature regards the dry solid as the stable state and the solution as the fragile one: refrigeration at 2-8 degrees C is standard for short holding periods, and minus 20 degrees C or colder for long-term storage. Vials are brought to room temperature before opening so that atmospheric moisture does not condense onto the cold solid. Engineered IGF-I variants are larger and more structurally complex than the short peptides in this catalogue; the formulation literature identifies aggregation on freeze-thaw as the principal handling risk.

For reconstitution, the diluent is added slowly down the inner wall of the vial rather than directly onto the cake, and the vial is swirled rather than shaken - agitation at an air-liquid interface is a well-documented driver of peptide aggregation. The solid should dissolve to a clear, particle-free solution; persistent cloudiness or visible particulate indicates the material should not be used.

Reconstituted solution is held at 2-8 degrees C and protected from light. Freeze-thaw cycling is the single most avoidable cause of loss, so where a solution must be frozen it is aliquoted first into single-use volumes. Working aliquots are labelled with the lot number so that any result can be traced back to the certificate of analysis for that lot.
