Mechanism of Action

This is a consumable kit with no pharmacological mechanism of its own. Its function is to hold a reconstituted peptide solution in a metered-spray container for intranasal delivery in a research setting.

Nose-to-brain delivery has been reviewed in Pharmaceutics and Bioorganic and Medicinal Chemistry, with attention to why the nasal epithelium is studied as a route that partly bypasses the blood-brain barrier and why bioavailability by that route is variable and formulation-dependent.

Research Findings

Indexed work covers nose-to-brain delivery of therapeutic peptides as nasal aerosols, nanoparticle and liposomal carriers, and microneedle-mediated approaches. Reported endpoints are pharmacokinetic and biodistribution measures in animal models.

Storage & Reconstitution

Components are stored sealed at controlled room temperature until assembly, protected from light. The bottle, actuator and supplied diluent are single-use for a given preparation; the assembled device is not disassembled, cleaned and refilled with a second preparation.

When preparing the solution, the diluent is added to the peptide vial rather than the reverse, down the vial wall and with swirling rather than shaking. The resulting solution is transferred once, into the spray bottle, and the number of transfer steps is kept to a minimum - each one costs material to adsorption and adds a contamination opportunity.

The assembled device is stored upright at 2-8 degrees C and protected from light. The kit should be labelled with the peptide identity, the lot number and the date of assembly so that any observation can be traced to a certificate of analysis.
